작용기전

콜레스테롤 흡수와 연관된 ezetimibe의 작용기전

Ezetimibe selectively inhibits absorption of cholesterol at the brush border membrane in the intestinal lumen. Interruption of cholesterol absorption stimulates LDL-receptor expression in the liver and thereby increases LDL clearance from the circulation.

peroxisome proliferator-activated receptor α(PPARα)에 결합, 활성화 → PPARα은 retinoid X 수용체(RXR)와 이질이합체 형성 → PPREs에 결합 → 유전자 전사↑, 단백질 발현↑ → 지질대사에 대한 영향: 혈장 중성지방농도↓, HDL↑

과산화소체 증식인자 반응요소(PPREs): 간세포, 골격근, 포식세포, 심장에 발현하는 핵 수용체인 과산화소체 증식인자에 의해 활성화되는 수용체 특정 유전자의 촉진자 (promoter)에 존재
